Rabbit peritoneal exudate cells (PEC) were exposed in vitro to Keyhole Limpet hemocyanin (KLH) antigen before or after exposure to 550 R X-irradiation. The ability of these cells to invoke an anti-KLH immune response was then studied in normal rabbits to which these cells were transferred. Irradiation of these cells before exposure to KLH abolished their capacity to provoke a specific antibody response; but irradiation after antigen exposure did not. These results indicate that irradiation can impair the ability of PEC to bind antigen but not their ability to migrate to or complete required interactions with immunocompetent lymphoid cells in the recipient rabbit. This effect was not apparent, however, when another antigen, bovine serum albumin, was used.
